摘要
目的:确定人胎儿骨髓间充质干细胞(Fetalmesenchymalstemcells,FMSCs)体外分离和培养的方法,观察其生物学特性,建立稳定的MSCs体外培养扩增体系。方法:通过密度梯度离心、贴壁筛选法分离培养人FMSCs,观察细胞的形态与超微结构,并研究其增殖及生长特征,应用流式细胞术测定细胞周期和CD14、CD29、CD34、CD44、CD45的表达。结果:在体外培养条件下人FMSCs贴壁生长,为成纤维细胞样,CD14/CD34/CD45阴性,CD29/CD44阳性,核浆比大,细胞周期检测G0/G1期约占95%,具有原始细胞的特征。结论:体外培养可获得较均一的FMSCs,形态单一、生长稳定、增殖性较强,可以用于细胞治疗的进一步研究。
Objective:To study the culture of fetal mesenchymal stem cells(FMSCs)- in vitro- from human fetal bone marrow and investigate their biological properties. To establish a steady culture system of MSCs for future study and clinical applications. Methods:FMSCs were obtained from fetal bone marrow by density gradient centrifugation and selected by cell attachment. The morphology and ultrastructure of FMSCs were observed by electron microscope. The cell cycle and surface antigens were analyzed by flow cytometry. Results:FMSCs were fibroblast-like cells. They were positive for CD29 and CD44, but were negative for CD14, CD34 and CD45. The nucleocytoplasmic ratio was high. About 95% of FMSCs were in G 0/G 1 phase and FMSCs had some primordial characteristics. Conclusion:Our results suggest that FMSCs could be of great interest for future application of cell therapy.
