摘要
目的:观察TGF-β1与PCNA在人胎儿睾丸发生过程中的表达特征,探讨二者在睾丸发生中的作用。方法:采用SP免疫细胞化学技术检测12~28周人胎儿睾丸间质细胞、支持细胞、生精细胞TGF-β1、PCNA的表达情况。结果:TGF-β1阳性细胞胞浆呈桔黄色或棕黄色颗粒状;在12周胎儿睾丸组织未见表达,在16~28周睾丸间质细胞呈阳性表达,16~20周为表达高峰;随着胎龄的增长TGF-β1阳性细胞率逐渐下降(P<0.01)。PCNA阳性细胞核为桔黄色或棕黄色,在胎儿睾丸间质细胞、支持细胞和生精细胞均有不同程度表达,16~20周为表达高峰,随着胎龄的增长PCNA阳性细胞率逐渐下降(P<0.01)。结论:TGF-β1与PCNA在人胎儿睾丸发育过程中有不同程度的表达,表明TGF-β1与PCNA在睾丸发育过程中起着一定的作用。
Objective: To observe the expression of TGF-β1, PCNA and study their roles during the development of human fetal testis. Methods:SP immunocytochemical technique was used to observe the expression of TGF-β1, PCNA in Leydig cells, Sertoli cells and spermatogenic cells of human fetal testis from 12th to 28th week.Results: The TGF-β1 immunopositive productions were orange or brown granules and scattered in cytoplasm, the positive cells were not found in 12th week testis, but were found in Leydig cells from 16th to 28th week. The peak expression was in 16-20th week and the number of positive cells decreased gradully with the increasing of fetal ages (P<0.01). The PCNA positive nuclei were orange or brown and the expression were observed differently in degree in Leydig cells, Sertoli cells and spermatogenic cells. The peak expression was in 16-20th week and the number of positive cells decreased gradually with the increasing of fetal ages (P<0.01).Conclusions: The expression of TGF-β1 and PCNA in different degree during the development of human fetal testis indicate that they might play important roles in the development of testis.
