摘要
通过使用几种不同溶剂对丹参进行提取,研究各提取物对酪氨酸酶的激活作用。结果表明:用乙醚作溶剂提取丹参得到的提取物对酪氨酸酶的激活效果较好(激活率129 0%),但提取率很低(10%左右)。用提取率较高的甲醇为提取剂,并对其浓缩物用极性由小到大的溶剂再进行萃取,并测定各萃取物对酪氨酸酶的激活率,得到乙醚萃取物对酪氨酸酶的激活率最高,且提取率远高于丹参的乙醚直接提取物。对该部分再经过硅胶柱分离,得到其中的一个甲醇洗脱液对酪氨酸酶激活率约是丹参甲醇提取物的3 5倍。
<Abstrcat>Danshen was extracted by using various solvents and the activating effect of every extract to tyrosinase was compared. Results indicate that the ether extract has the highest activating effect to tyrosinase, but the yield is low.Then the solvent with high yield, methanol was used for the extraction and the methanol extract was further extracted by a series of solvent of different polarity, from lower to higher respectively. The extracts were measured for activating effect to tyrosinase. It was concluded that the ether extract has the highest activating effect while the yield is significantly higher than that obtained by direct extraction with ether. Then the ether extract was separated by using a silica gel column and one of the methanol elution from the silica gel column achieves activating effect of 3.5 times that of the methanol extract.
