摘要
目的:探讨脑出血后脑组织c-fos表达和一氧化氮合酶(NOS)阳性神经元活性的变化,观察下瘀清痰汤对这种变化的影响.方法:采大鼠自体血注入脑内制成脑出血模型,造模后分批采集标本,用免疫组化法观察c-fos的蛋白表达,用NADPH-d组化法测定NOS阳性神经元的分布,用双重染色技术观察fos/NOS双标细胞的变化,分析下瘀清痰汤的干预结果.结果:造模后1 h,脑组织即有fos出现和NOS活性的增强;造模后3 h fos表达达高峰;6 h NOS阳性神经元活性增强、数目增多达高峰;约10%~15%的NOS阳性神经元同时呈现fos表达阳性.而以下瘀清痰汤干预的中药组与模型组相比,不同时间点NOS阳性神经元数目与fos表达以及NOS/fos双染细胞数均明显低下.结论:c-fos和NOS参与了脑出血的病变过程,而下瘀清痰汤治疗脑出血的机制可能与其抑制c-fos表达、减弱NOS阳性神经元的活性有关.
Objective:to study the expression of c-fos and the activity of NOS in rats brain tissue after cerebral hemorrhage,by observing the effects of xiayu qingtan decoction.Methods:inject self-blood into rats′caudatoputamen to make cerebral hemorrhage models,and collect various targets in batch.Observe the expression of c-fos by means of immunity histochemistry,measure the distribution of NOS positive neuron by using NADPH-d histochemistry,observe the distributions of double labeled fos/NOS cells with double staining technique.Results:fos appeared in rats brain tissue only one hour after cerebral hemorrhage model was made,and reached its peak 3 hours later.The activity of NOS in rats brain tissue was intensified and reached its peak in 6 hours.Among the NOS positive neurons,about 10%~15% of them were fos positive.Compared with the model group,the xiayu qingtan decoction group had lesser numbers of NOS positive neuron,lower expression of fos,and lesser numbers of double labeled fos/NOS neurons.Conclusion:c-fos and NOS participated in pathologic change of rats cerebral hemorrhage.The effects of xiayu qingtan decoction on cerebral hemorrhage may related to restraining the expression of c-fos and reducing the activity of NOS in rats brain tissue after cerebral hemorrhage.
