摘要
目的了解血管形成因子-1(Angpt-1)与肺动脉高压(PH)形成的关系。方法收集11例存在PH的先天性心脏病(先心病)患儿的肺组织,应用荧光定量PCR法分析Angpt-1mRNA的表达率,免疫组化技术分析Angpt-1蛋白的表达;并将11例先心病伴肺高血流患儿和6例正常儿童肺组织进行比较。结果PH患儿和肺高血流组患儿肺组织中Angpt-1mRNA的表达率和Angpt-1蛋白的表达均高于正常组(P<0.01)。结论Angpt-1参与和促进先心病肺动脉高压的发生和发展。
Objective To explore the changes of angiopoietin-1 (Angpt-1) level in the lung tissues in childrenwith congenital heart disease (CHD) complicating pulmonary arterial hypertension (PAH). Methods This study enrolled two groups of the patients(11 patients with CHD complicating PAH(Group Ⅰ)and 11 patients with CHD with high blood stream(Group Ⅱ)) . The expressions of Angpt-1 in the lung tissues were investigated with reverse transcriptase-polymerase chain reaction(RT-PCR) assays and immunohistochemical analyses in comparison with the results from six control cases. Results RT-PCR quantitative assays revealed that Angpt-1 mRNA expression was increased in Group Ⅰ and Group Ⅱ (P<0.01) .Whereas, immunohistochemical analyses revealed that there was strong immunostaining for Angpt-1 in Group I and Group Ⅱ. Conclusions Angpt-1 participated and accelerated the development of PAH in the patients with CHD.
