摘要
应用纯化的D11^4株抗马立克氏病病毒中和性单克隆抗体,以SPA-IgG法和脂质体-IgG法制成免疫原,分别免疫BALB/c小鼠,应用B淋巴细胞杂交瘤技术进行细胞融合,以两步ELISA筛选法获得了3株抗独特型单克隆抗体,分别命名为1B8、10C1和3D5,其Ig亚类分别为IgG1,IgG1和IgG2b,染色体数在92-102之间,在捕获阻断ELISA中。
Three anti-idiotypic antibodies (McAb,) named 1B810C1 and 3D5 were developed by fusion of mouse myeloma cell line NS-1 and spleen cells of BALB/c mice immunized with immunogens prepared from purified D114 neutralizing monoclonal antibodies to MDV and SPA or liposome respectively. The antibodies secreted by these hy-bridomas belong to IgG1 and IgG2 respectively. Binding activity of MDV antigen and McAb1 was specifically blocked by the McAb2 from mouse ascitic fluid in catcd-blockingELISA, and the blocking rate was 2. 66% to 76. 4% in the 1 :20 to 1:80 000 diluted McAb2 samples. The purified McAb2 which were emulsified with an equal volume of oil adjuvant were injected into 20-day-old chickens for three times and the serum samples were taken 10 days after final immunization and were tested for the presence of antibodies to MDV antigens. The results showed that the specific antibodies were present in the serum samples of immunized chickens. It was suggested that the configulafion of the variable regions of the McAb2s may be similar to those of antigenic determinant of MDV which combine with McAb1. The McAb2s will be useful in the research on monoclonal anti-idiotypic vaccine for Marek's disease.
