摘要
目的获得不同转移潜能的骨肉瘤细胞株。方法从我室建立的人成骨肉瘤细胞系SOSP-9607中通过有限稀释法获得不同的单细胞克隆,经过侵袭试验初筛后获得5株转移潜能不同的细胞株,再通过裸鼠体内连续传代法筛选出具有不同转移能力的细胞系。结果获得两株转移潜能不同的骨肉瘤细胞株。结论这两株细胞株它们来源于同一亲本,他们之间的差异集中在转移表型上,为今后利用分子生物学的方法筛选出转移相关基因,从而为成骨肉瘤的转移机制及临床研究奠定基础。
Objectives To get the the human osteosarcoma cell sublines with different potential of metastasis. Metheds The different monocell clones were obtained by the limited dilution method from the human osteosarcoma cell line,sosp- 9607. After preliminary screening out by in vitro invision assay,we selected two cell lines as the high metastasis potential cell sublines and three cell lines as the low metastasis potential cell sublines.Then, the cells lines with different poentials of metastasis were screened out by a serial passage in the mice. Results The two sublines with the different metastasis potential were gotten. Conclusions These two subh'nes came from the same mother cell line. The main difference between them lies in the different metastatic phenotype. This study provides an base for the screening of the gene correlated with metastasis and even for the further metastatic mechanism and clinical study of osteosaroma.
