摘要
目的构建反义及正义波形蛋白重组逆转录病毒载体,研究波形蛋白在反应性胶质化中的功能与作用。方法应用分子克隆方法克隆、鉴定、包装并感染培养的星形胶质细胞。结果同时获得全长反义及正义Vim逆转录病毒克隆细胞株和较高滴度的病毒上清,反义Vim逆转录病毒抑制培养星形细胞的Vim表达。结论重组的反义及正义Vim逆转录病毒可用于进一步的实验研究。
Objective To construct antisense and sense vimentin retrovirus vector for investigating the effect of vimentin on reactive gliosis. Methods Molecular clone methods were used for tp clone,identifing the antisense and sense vimentin retrovirus and their package.The astrocytes were infected by retrovirus,cultured and determined by SABC method. Results Both the antisense and sense vimentin retrovirus were gained and also high titer of retrovirus superfactant was gotten. The expression of vimentin in the cultured astrocytes was inhibited by antisense vimentin retrovirus compared with sense retrovirus. Conclusion Antisense and sense vimentin retrovirus can be used for the study of reactive gliosis.
