摘要
目的 研究人脑出血后海马神经元损伤与凋亡促进因子Caspase- 3之间的关系。方法 选取10例因脑出血而死亡的尸检脑标本,应用HE染色,Caspase- 3免疫组织化学染色和Caspase- 3m RNA原位杂交,观察脑出血时海马CA1 区神经元损伤变化与Caspase- 3表达之间的关系。结果 海马CA1 区,出血5 h可检测Caspase- 3免疫组化染色的阳性细胞(10 .4个/高倍视野) ,2 2~4 8h时达高峰(2 1.7~2 4 .3个/高倍视野)。Caspase- 3m RNA原位杂交阳性表达始于5 h(6 .75个/高倍视野) ,2 4 h达高峰(14 .6 0~18.30个/高倍视野) ;二者均在72 h后呈明显下降趋势。出血2 4 h,可检测到TUNEL阳性细胞,持续至72 h。Caspase- 3m RNA与Caspase- 3呈正相关,相关系数为0 .717(P<0 .0 5 )。4~16 h受损神经元形态基本正常,2 4~4 8h细胞凋亡特征明显,72 h后,几乎所有神经元形态呈现严重病理变化。结论 人脑出血后海马神经元发生一系列形态学变化,其演变规律与凋亡促进因子Caspase- 3有密切相关性。
Objective Acute hemorrhagic stroke is one of the most devastating types of stroke,and which is known to cause severe neuronal damage.This study was undertaken to investigate the relationship between hippocampus neuronal injury and expression of Caspase-3 after acute hemorrhagic stroke.Methods We performed post mortem analysis of brain hippocampus CA 1 area in 10 patients who had died from acute hemorrhagic stroke.Histopathologic changes were observed by HE stain, and Caspase-3 protein and mRNA expression level was assessed by immunohistochemistry and in situ hybridization respectively.Results Caspase-3 protein immunohistochemical staining positive cells(10.4/HP) were identified from 5h after acute hemorrhagic stroke in hippocampus CA 1 area,and reached the peak at 22~48h(21.7~24.3 /HP).Caspase-3 mRNA in situ hybridizational positive expression cells were identified from 5h after hemorrhagic stroke (6.75/HP),which reached the peak at 24h (14.60~18.30/HP).Caspase-3 mRNA and protein expression level was decreased after 72h.There was a statistically significant correlation between Caspase-3 mRNA and protein expression(r=0.717,P<0.05).TUNEL-positive cells appeared from 24h to 72h after hemorrhagic stroke.The injuried neurons remained normal morphology in 4~16h,and the features of cellular apoptosis showed apparently in 24~48h.after 72h,almost all of the neurons presented severe histopathologic changes.Conclusion Our data demonstrated a series of morphologic alteration in hippocampus neurons after acute hemorrhagic stroke,which is consistent with the expression of Caspase-3 mRNA and protein.
