摘要
目的 研究氯乙烯染毒大鼠肝微粒细胞色素P4502E1(CYP2E1)活力和mRNA表达随染毒时间和剂量的变化, 探讨两者可能的关系。方法 大鼠染毒12 周, 低、中、高剂量组的染毒剂量分别为5,10, 20 mg/kg, 分别在第3, 6, 9 和12 周处死动物, 代谢酶CYP2E1 活力测定采用分光光度法, 其mRNA表达用一步法RT PCR测定。结果 低剂量组CYP2E1活力随染毒时间增加而增强(P<0 05);中、高剂量组CYP2E1活力出现先升高后降低的趋势。染毒结束时,中、高剂量组大鼠肝细胞mRNA表达明显高于对照组(P<0 01)和低剂量组(P<0 05)。结论 低剂量组CYP2E1 的活力随染毒时间增加而增强, 而CYP2E1 mRNA的相对表达在本次实验中未出现时间趋势, 但在染毒结束时有明显的剂量-效应关系, 两者变化水平不一致。
Objective To investigate the level of activity and mRNA expression of CYP2E1 in vinyl chloride monomer(VCM)exposed rats and to explore the possible mechanism of VCM induced liver damage.Methods Doses of 0,5,10, 20 mg/kg VCM were respectively administered to animals for 12 weeks by ip,which were sacrificed at the 3 rd ,6 th ,9 th ,and 12 th week.The activity of CYP2E1 in liver was determined using spectrophotometry.The expression of hepatic CYP2E1 mRNA was measured by one step RT-PCR.Results The activity of hepatic CYP2E1 in 5 mg/kg group increased as the experiment progressing (P<0.05) ,while this index increased followed with its decreasing in the 10,20 mg/kg groups.At the end of experiment,CYP2E1 mRNA levels of 10,20 mg/kg groups were significantly higher than control and 5 mg/kg group,P<0.01 and P<0.05 respectively.Conclusions The activity of CYP2E1 in 5 mg/kg group increased during the administration.CYP2E1 mRNA content didnt correspond to its activity;the former may be better in describing the model of dose-response.
