摘要
本文应用放射性同位素前体参入法研究多抗甲素对人慢性髓性白血病细胞株K562,人早幼粒白血病细胞株HL-60,鼠X鼠杂交瘤细胞株E4B11C9和小鼠黑色素瘤细胞B16-F10的DNA,RNA和蛋白质合成作用的影响。结果表明:10~5000μg·ml-1的多抗甲素体外对四株肿瘤细胞的DNA,RNA和蛋白质合成均有不同程度的抑制作用,并存在明显的剂量依赖性和瘤株差异性。在大剂量时以对RNA的抑制作用最强。本实验为多抗甲素的抗肿瘤作用机制研究及临床应用提供了有益的实验依据。
Polyactin A(PAA),a new home-tnade biological respon-se modifier,has various biological activities,The precursors of radioac-tivity uptake assay,which measure the incorporation of 3H-TdR or 3H-UR or 3H-Leu into DNA,RNA or protein by surviving cells,were used to study the effect of PAA on K562,HL-60,E4B11C9 hybridoma and B16-F10 melanoma cell in vitro,It was found that PAA ranged from 10 to 5000μg/ml could inhibited the DNA,RNA and protein synthesis in the K562,HL-60,E4B11C9 hybridoma and B16- F10 melanoma cell at different degree,Especially,the RNA synthesis was strong inhibited at large dose.The effect of PAA was not only dose-dependent,but also different from the cell lines,These results suggest that PAA possessed the inhibitory actions of cell growth,which may play an important role in antitumor activity.
