^(131)I-血管抑素对小鼠Lewis肺癌移植瘤的干预效应

摘要 目的:观察131I标记的血管抑素在荷Lewis肺癌C57BL/6小鼠体内的放射受体显像,评价血管抑素及131I-血管抑素和单纯131I对小鼠Lewis肺癌移植瘤的治疗效果,探讨131I-血管抑素治疗实体瘤的临床应用前景。方法:①2001-02/05在第四军医大学唐都医院核医学实验室进行显像实验:用氯胺-T法进行131I标记,将标记产物131I-血管抑素从尾静脉注入荷Lewis肺癌的C57BL/6小鼠体内,用131I标记的白蛋白作为对照,分别于24,48,96,144h后进行ECT扫描。②2002-07/2003-02在该实验室进行治疗实验:28只荷Lewis肺癌的C57BL/6小鼠(右前肢皮下移植瘤直径约1cm)随机分成4组,分别腹腔注射131I-血管抑素(含131I11.1MBq,血管抑素12.5mg/kg),血管抑素(12.5mg/kg),131I(11.1MBq)和生理盐水各0.3mL,1次/周,治疗2次,观察14d内肿瘤体积的变化。结果:131I-血管抑素尾静脉注射后,48h内γ显像以全身分布为主,96h后肿瘤部位显像,并呈高度特异性浓集。131I-血管抑素,131I,血管抑素和生理盐水治疗两周后,各组小鼠Lewis肺癌移植瘤的平均体积分别为(1963±126),(5219±351),(3943±236),(7353±350)mm3,其抑瘤率分别为73.3%,29.0%,46.4%,与生理盐水组比较,差异有显著性意义(P=0.0003)。结论:131I-血管抑素可以高度特异性浓集于小鼠Lewis肺癌移植瘤部位, AIM:To observe the radio receptor imaging of 131I angiostatin in C57BL/6 mic e bearing Lewis lung cancer,evaluate the therapeutic effects of angiostatin,131I angiostatin and 131I on transplanted Lewis lung carcinoma in mice and investig ate the clinical prospect of 131I angiostatin for solid tumor. METHODS:The imaging experiment was conducted in the laboratory for Nuclear Med icine,Tangdu Hospital,Fourth Military Medical University of Chinese PLA between February and May 2001.①Angiostatin was labeled with 131I by the conventional ch loramines T(Ch T) method.131I angiostatin was injected into C57BL/6 mice throu gh caudal vein whose forelimbs were transplanted with Lewis lung carcinoma,and 1 31I labeled albumin(131I Alb) as control.The whole body imaging was acquired a t 24,48,96 and 144 hours after injection.②The treatment was conducted in the sa me laboratory as above from July 2002 to February 2003.Twenty eight male C57BL/ 6 mice(the mean diameter of tumors transplanted into the right forelimbs was abo ut 1 cm) whose forelimbs were transplanted with Lewis lung carcinoma were divide d into four groups.Tumors in the four groups were treated with 131I angiostatin (131I 11.1 MBq per mouse,angiostatin 12.5 mg/kg),angiostatin(12.5 mg/kg),131I(11 .1 MBq per mouse) respectively and with similar normal saline(NS) 0.3 mL as cont rol.Each drug was given intraperitoneally and injected for two times at an inter val of 7 days respectively.The volume of tumors was measured during 14 days afte r treatment. RESULTS:After injection of 131I angiostatin,131I angiostatin was distributed in whole body of mice during 48 hours of γimaging,and the tumor showed clearly at 96 hours after injection at a highly specific concentration.The mean volume of transplanted tumors in the mice with Lewis lung cancer was(1 963±126),(5 219 ±351),(3 943±236) and (7 353±350) mm3 after treated with 131I angiostatin,13 1I,angiostatin and NS respectively,and the growth of transplanted Lewis lung car cinoma was inhibited by 73.3%,46.4%,and 29.0%in the treatment groups respecti vely,significantly different from that in the control group(P=0.000 3). CONCLUSION:131I angiostatin can highly and specifically concentrate in the tu mor transplanted with Lewis lung carcinoma and can show the imaging of tumor tis sues;It can also inhibit the growth of transplanted tumor of Lewis lung carcinom a in mice,and its inhibitive effect is better than that of 131I or angiostatin.S o it is suggested that 131I angiostatin has potential prospect of clinical appl ication in the treatment of solid tumor
