摘要
目的:研究大鼠在脑室埋管注射甘珀酸预处理后戊四氮导致癫痫发作时前脑内星形胶质细胞和连接蛋白43的形态学反应及其相互关系。方法:实验于2003-10/2004-06在解放军第四军医大学神经科学研究所进行。取24只大鼠分为生理盐水组、甘珀酸组、戊四氮组、甘珀酸+戊四氮组,应用免疫荧光组织化学双重标记显示胶质纤维酸性蛋白和连接蛋白43蛋白在前脑的表达及其相互关系。结果:甘珀酸+戊四氮组的大鼠癫痫发作的行为表现比戊四氮组显著加重,星形胶质细胞胶质纤维酸性蛋白的表达也比戊四氮组明显升高,值得注意的是甘珀酸组的星形胶质细胞胶质纤维酸性蛋白的表达比生理盐水组明显升高。连接蛋白43在戊四氮引起的癫痫大鼠的大脑皮质、海马和杏仁核内增加,而甘珀酸预处理后的癫痫模型中连接蛋白43进一步增强。在连接蛋白43与胶质纤维酸性蛋白双标的切片上发现,阳性的星形胶质细胞双标,在甘珀酸预处理后的癫痫模型中胶质纤维酸性蛋白阳性的星形胶质细胞显著增加的同时连接蛋白43也明显增加。结论:在整体动物甘珀酸预处理可以导致癫痫发作增强,这可能与星形胶质细胞增生及其连接蛋白43表达升高有关。
AIM:To observe the morphological response of astrocytes and connexin 43(Cx43) and their relationship with each other in forebrain of pentylenetetrazol(PTZ) induced seizure rats pretreated with carbenoxolone(CBX). METHODS:The experiment was finished in the Institute of Neurology,Fourth Military Medical University of Chinese PLA from October 2003 to June 2004.Twenty four rats were divided into saline group(S group),CBX group,PTZ induced seizure group(PTZ group) and CBX+PTZ group.The double labeled fluorescence immunohistochemical staining for glial fibrillary acidic protein(GFAP) and CX43 was used to investigate the expression of GFAP positive cells and CX43 protein and their relation. RESULTS:The behavior of seizure in CBX+PTZ group was more serious than that in PTZ group,and the expression of GFAP in CBX+PTZ group was also obviously increased.Noticeably,the expression of GFAP in CBX group was much higher than that in NS group.The distribution of Cx43 in cortex,hippocampus and amygdaloid nucleus was increased in PTZ induced epilepsy rats,and Cx43 was farther increased in CBX+PTZ group.On the slices with Cx43 and GFAP double labeled,the Cx43 and GFAP double staining positive astrocytes were observed.The GFAP positive astrocytes increased obviously,and the expression of Cx43 also increased obviously in CBX+PTZ group. CONCLUSION:The seizure can be enhanced by CBX pretreatment and it may relate with astrocytic hyperplasia and the increasing expression of Cx43 in astrocytes.
