超抗原金黄色葡萄球菌肠毒素A对异位皮炎患者外周血单个核细胞活化作用的观察 被引量:1

摘要 目的观察超抗原金黄色葡萄球菌肠毒素A(SEA)对异位皮炎(AD)患者外周血单个核细胞(PBMC)的活化作用。方法用不同浓度的SEA刺激AD患者PBMC,MTT法观察PBMC增殖及流式细胞术检测T细胞表型情况。结果AD患者皮损表面金葡菌检出率为91.2%;当SEA为0.8μg/ml时PBMC表现了最大的增殖活性,健康志愿者SEA刺激后T细胞CD3、CD4、CD8表达显著降低,AD患者SEA刺激后T细胞CD3、CD4、CD8的表达显著增高,AD患者和健康志愿者SEA刺激前后CD4/CD8差异无显著性。结论AD患者皮损对金葡菌具有亲合性,SEA与AD发病有关。 Objective The purpose of the experiment is to ob serve the activation of AD patient PBMC with superantigen SEA .Methods MTT assay was used to detect the proliferation of healthy people and AD patient PBMC with varient concentration of superantigen SEA; Flow cytometry(FCM) was used to detect the activation of CD_3+,CD_4+,CD_8+ T cells with SEA in culture conditions.Results The inspected rate of S. aureus on the focus of AD patient was 91.2%; 0.8% concentration of SEA had strongest ability to stimulate PBMC to proliferat e. The expression of CD_3, CD_4, CD_8 decreased in healthy people with SEA in c ulture conditions and increased in AD patient after the stimulate of SEA, the differen ce of CD_3, CD_4, CD_8 between healthy people and AD patients was prominent. SE A showed the same activation to CD_4, CD_8 T cells in culture conditions ,the expressi on of CD_4/CD_8 in healthy people and AD patients did not show any statistics difference after the stimulate of SEA.Conclusion The inspected rate of S. aurcus on the focus of AD patient was high; SEA is related with the pathogcncsis of at opic dermatitis
