摘要 目的:观察模拟微重力旋转培养系统(RCCS)对骨髓间充质干细胞(mesenchymal stem cells,MSCs)体外培养的影响。方法:将兔MSCs与三维载体PGA相复合后,置于RCCS系统中进行培养,观察MSCs增殖能力和超微结构的变化。结果:经过RCCS系统培养,MSCs增殖能力明显增强,细胞形态较静置培养组更接近生理情况。结论:采用RCCS系统进行细胞/载体结合物的体外培养,增殖能力有明显增强,细胞的形态更接近生理状态,有助于培养出接近正常软骨生物学特性的工程化软骨。 Objective: To investigate mesenchymal stem cells(MSCs) proliferation under microgravity rotating culture system. Methods: Expanded and combined with PGA, MSCs were cultured under the microgravity rotating cell culture system(RCCS).The proliferation of MSCs was evaluated by MTT assay, and the biological features of the compound were also observed under the scanning electron microscope. Results: The MSCs′ proliferation was enhanced markedly,the cells′ morphology were near to those under physiology state.Conclusion:Cultured under the RCCS, the proliferation of MSCs was enhanced markedly. And the amelioration of the MSCs′ biological features was also found by the scanning electron microscope.
