NM-3诱导裸鼠体内人胃癌细胞的凋亡机理 被引量:2

Mechanism of Apoptosis of Human Gastric Cancer Cells Induced by NM-3 in Nude Mice

摘要 目的:探讨新的血管生长抑制剂NM- 3对体内人胃癌细胞凋亡的作用。方法:建立人胃腺癌裸鼠皮下异体移植模型。移植后第1周开始腹腔注射NM- 3,1周3次,剂量为10mg/kg、20mg/kg、40mg/kg,分别联合卡铂5mg/kg,1周2次;并与生理盐水对照组,NM -3或卡铂单独治疗比较(应用NM 3 5 周,卡铂3 周)。移植后第7 周处死裸鼠,测量肿瘤大小,计算抑瘤率,FCM法分析肿瘤细胞凋亡指数(AI)。结果:单用NM- 3剂量分别为10mg/kg、20mg/kg及40mg/kg时,肿瘤的平均重量分别为1220±116.85mg、1041±143.51mg和767.5±140.11 mg,显著低于生理盐水对照组的肿瘤平均重量1754±144.18mg(P<0.05)。单用卡铂组的抑瘤率为5.6%,对人胃癌没有明显抑制作用。当单用NM 3 剂量10 mg/kg、20 mg/kg、40 mg/kg时,抑瘤率分别为30.4%、40.6%、56.2%,相对于对照组有明显的抑瘤作用(P<0.05),而且显著高于卡铂组抑瘤率5.6%。NM 3 3个剂量组联合卡铂治疗的AI分别是:5.66%±0.65%、8.98%±1.43%、21.66%±2.96%;生理盐水对照组为1.37±0.19%,卡铂组为1.85±0.22%。结论:NM 3能诱导体内人胃癌细胞的凋亡,抑制胃癌的生长。 Objective: To investigate the effects of a novel angiogenesis inhibitor, NM-3,on primary tumor growth and metastasis of gastric cancer in vivo.Methods: we established subcutaneous xenotransplanted model of a human gastric cancer, SGC-7901, by fixing human tumor tissue under the dorsal skin of nude mice. NM-3 was administered i.p. at a dose of 10mg/kg、20mg/kg and 40mg/kg, three times a week for 5 weeks,combined with carboplatin (5mg/kg) twice a week for 3 weeks. Compared to the combined treatment, the mice of other groups were injected with NM-3/carboplatin alone. Seven weeks after implantation, tumer size、tumor growh inhibition rate and apoptotic index were obtained after the mice were sacrificed. Results: compared with the untreated controls, the size of subcutaneously implanted tumor was significantly reduced in mice treated with NM-3 (10mg/kg, 20mg/kg, and 40mg/kg) with an inhibition rate of 30.4%, 40.6%, 56.2%, respectively. The AI was increased significantly in treated mice with NM-3 combined with carboplatin( 5.66± 0.65)%、( 8.98± 1.43)%、( 21.66± 2.96)% versus( 1.37± 0.19)%. Conclusion:Angiogenesis inhibitor NM-3 can induce apoptosis of human gastric cancer cells and has strong inhibitory effect on tumor growth of human gastric cancer in nude mice.
