摘要
将胸腺细胞分为正常对照组、H2O2模型组、0.1g/LVc组、0.5g/LPCF组、0.25g/LPCF组和0.125g/LPCF组6组,通过不同药物或不同浓度处理后,采用生物化学方法分别检测细胞裂解液中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、总抗氧化能力(T-AOC)和活性氧(ROS)的含量,采用流式细胞仪测定各组细胞胞浆中的Ca2+含量和线粒体膜电位(ΔΨm)的变化。结果显示,扇贝多肽(PCF)能提高细胞中SOD,GSH-Px的含量,增强细胞的T-AOC,降低细胞中MDA和ROS的含量,并能引起胞浆中Ca2+含量的下降和ΔΨm的提升。表明PCF具有抑制H2O2对胸腺细胞氧化损伤的作用。
<Abstrcat>To study the protective effects of polypeptide from Chlamys farreri (PCF) on thymocytes damaged by H_2O_2 and antioxidative mechanisms of PCF,we made a thymocytes damaged-H_2O_2 model.In this study,thymocytes were divided into six groups including control group,model group,0.1 g/L Vc group,0.5 g/L PCF group,0.25 g/L PCF group and 0.125 g/L PCF group.Thymocytes in different groups were exposed to PCF at various concentrations and 12.5 μmol/L H_2O_2.Levels of superoxide dismutase (SOD),glutathione peroxidases (GSH-Px),Total Antioxidative Capacity (T-AOC),malondialdehyde (MDA) and reactive oxygen species (ROS) in cell lysates were estimated by biochemical methods.Cytosolic Ca^(2+) levels and mitochondria membrane potential (ΔΨm) were determined by flow cytometry.Results showed PCF could enhance the activities of SOD and GSH-Px,promote antioxidative capacity and reduce the amount of MDA and ROS.Moreover,PCF caused an elevation of ΔΨm and a decrease of cytosolic Ca^(2+).It is concluded that PCF can inhibit the damaging effect of H_2O_2.
