The Effect on Tumour Immunity of Hepatocarcinoma Cells Induced by IL-12、IFN-γ Synergizes CD80 Costimulation

摘要 目的观察IFN-γ、IL-12对转染CD80基因前后肝癌细胞表面分子CD80的表达,评价CD80分子联合细胞因子对肝癌细胞杀伤效应的影响。方法取HepG2细胞和经逆转录病毒载体介导CD80转染的HepG2/CD80细胞,分别经IFN-γ、IL-12处理后,流式细胞仪检测CDS0分子的表达水平。LDH释放试验检测刺激前后肝癌瘤苗对CTL杀伤效应的影响。结果HepG2细胞表面CD80分子表达水平低,转染后HepG2/CD80细胞的表达增高。经IL-12、IFN-γ细胞因子诱导后,HepG2/CD80细胞表面的CD80基因表达进一步增强。细胞因子联合CD80基因转染提高细胞毒性淋巴细胞对肝癌细胞的杀伤作用,与对照组比较有显著差异(P<0.05)。结论(1)IL-12、IFN-γ刺激转染CD80肝癌细胞共刺激信号的进一步表达。(2)CD80基因转染联合IL-12、IFN-γ明显增强CTL对肝癌细胞的杀伤作用,提示两者存在协同作用。 Objective To observe the expression of CD80 transfected liver cancer cells (HepG2/ CD80) induced by IL-2、 IFN-γ, and evaluate the effection of CD80 molecule combined with IFN-γ/ IL-12 in the primary cytolytic activity against HCC. Methods HepG2 cells and HepG2/CD80 transfected via retroreverviral transduction were collected, the respective expression of CD80 co stimulated molecules by flow cytometric(FCM) after treated with IFN-γ, IL-12 were observed. LDH releasing test was used to check the cytotoxic activity of CTL. Results FCM revealed that CD80 molecules were expressed on the HepG2 cell surface, however, the level of CD80 expressing was very low. CD80 molecule expressing on the HepG2/CD80 cell surface was evry high. Aftert treatment with IL-12 and IFN-γ, the expressing of CD80 on HepG2/CD80 cell surface increased furthermore. Meanwhile, contradistinguishing from the discrepancy of the counterpart group, the cytotoxic activity of CTL against HepG2/CD80 is greatly improved in the above group with two combinations (P<0.05). Conclusion (1) IL-12 and IFN-γ increase the CD80 expressing of the transfected CD80 hepatocellular carcinoma cells. (2)CD80 gene transfer combining with IL-12 and IFN-γ greatly improved the cytotoxic activity of CTL on liver cancer cell, and we draw a conclusion that there is association between them.
