摘要
为了研究间隙连接在肿瘤促进中的可能作用,本室新近发展了一种水晶体离体培养技术,我们利用这种技术进行了促癌剂TPA促进水晶体过度生长的实验研究,即把20期的鸡胚水晶体培养于含有80ng/ml TPA的DMEM中,39℃下培养24小时。实验结果指出,实验组的水晶体非常明显地大于对照组(P<0.01)。已报道这个浓度的TPA可使间隙连接通讯安全抑制,表明以上结果可能与细胞间通讯的被抑制密切相关。联系到我们过去利用单克隆抗体所作的实验研究,本文结果进一步证明了间隙连接在生长控制中起重要作用;间隙连接通讯的抑制可能正是肿瘤促进的一种细胞机制。此外,本文报道的鸡胚水晶体离体培养似乎也可作为检测有害物质对生长发育影响的实验系统。
A method for culturing chicken embryo lens in vitro was devLenses form right eyes of chicken embryo at the stage 20 were cultured with TP etrade canoyl phorbol-13 acetate) at the concentration of 80ng/ml in DMEM; for the control. lenses from left eyes were cultured in DMEM without TPA. Lenses were cultured in an incubator at 39℃ for 24 hours. The size of lens, including length and width, was measured under a dissecting microscope, and the paraffin sections of lens were made with routine method after fixation. Statistical analysis of measured data showed that the lens in TPA group is obviously larger than that in control (P<0.01) , but no significant difference in size (P>0.05) was detected by comparing lenses between right and left eyes of the same chicken embryo. It implicated that the overgrowth of lens is caused by the action of TPA. It was reported that TPA at the concentration of 10-100ng/ml induced almost complete inhibition of cell com mmunication and this inhibition was continued for at least 24 hours in the presence of TPA. This suggested that the overgrowth of lens is correlated with the inhibition of intercellular co mmunication . The result reported here is consistent with that of our previous work using monoclonal antibody against gap junction protein to block the formation of gap junction. These results indicated further that the gep junction plays an important role in growth control and the inhibition of gap junctional communication could be a possible cellular mechanism of tumor promotion.Any change in morphology was not observed and the lens still developed under our experimental condition. The method for culturling lens in vitro may be served as an useful model system to examine the effects of toxic chemicals on growth and development.
