兔骨髓基质干细胞体外培养的生物学特性

摘要 目的:分离培养兔骨髓基质干细胞并探讨其体外生物学特性。方法:通过梯度密度离心法分离兔骨髓基质干细胞,体外培养、扩增,观察细胞的大体形态和超微结构;通过绘制细胞生长曲线、MTT实验来研究细胞的增殖能力和代谢活性。结果:兔骨髓基质干细胞呈梭形,胞质内有丰富的粗面内质网和线粒体,具有很强的增殖能力和代谢活性。结论:本实验方法取材方便。 Objective: Isolated cultures of rabbit bone marrow mesenchymal stem cells (MSCs) were made to study their biological characteristics in vitro. Methods: MSCs were isolated from bone marrow by density gradient centrifugation. Then the MSCs were purified and cultured. The appearance of MSCs was observed under the phase contrast microscope. Microstructure of the MSCs was observed under transmission electron microscope. The cell growth curve was drawn and MTT assay was tested to study the reproductivity and metabolic activity of MSCs. Results: Under the phase contrast microscope, the appearance of the MSCs was spindle like, and there were a large number of mitochondrion and rough endoplasmic reticulum in the nucleus of MSCs. Conclusion: It's easy to obtain materials for the experiment in obtaining MSCs in a large number.;
