应用PCR检测环境水体中的嗜肺军团菌 被引量:4

摘要 目的 建立检测Mip基因的PCR方法,以检出环境水体中的嗜肺军团菌。 方法 采用Godkey软件设计引物,制备菌种模块,对模拟和环境水样进行处理,然后作PCR扩增及其产物检测。 结果 该方法检测军团菌的灵敏度为5 .8×10 2 cfu/ml,6株标准军团菌均扩增出65 0bp的条带,4株非嗜肺军团菌和4株非军团菌无条带出现;检测71份环境水样,5份阳性,阳性率为7.0 %。 结论 该方法快速、灵敏、特异。 Objective Mip gene Polymerase Chain Reaction (PCR ) assay was established to detect Legionella pneumophila in environmental waters. Results The sensitivity was 5.8×10 2 cfu/ml, 650 bp DNA fragment can be amplified reproducibally from 6 reference Legionella pneumophila strains, no similar DNA product was observed in 4 non-Legionella pneumophila strains and 4 non-Legionella strains. Five samples were obtained positive result in 71 examined samples, the positive rate was 7.0%. Conclusions This assay is highly sensitive and specific and offers an effective method to detect Legionella pneumophila in the environmental waters.
