摘要
目的建立从酸枣仁中提取白桦脂酸的方法,并进行质量控制。方法按工业化生产要求,用水和乙醇提取,用石油醚除去酸枣仁中的脂肪油,采用紫外扫描法(将提取物用甲醇稀释成20μg·mL1的溶液,在190~400nm波长范围内进行扫描)、红外扫描法(提取物用甲醇溶解后涂在岩盐窗上采用红外色谱扫描)和高效液相色谱法[色谱柱为ALCHROMTMHyperSilC18;流动相为甲醇∶纯化水∶三乙胺(92∶8∶0.04);流速为0.8mL·min1;检测波长:210nm,柱温:室温]对所获得的白桦脂酸进行定性和定量分析。结果从酸枣仁中提取出晶体后,经紫外、红外扫描法和高效液相色谱法定性为白桦脂酸。酸枣仁中白桦脂酸的含量约为0.084%;测得提取的白桦脂酸纯度为86.6%。结论用该法从酸枣仁中提取白桦脂酸,提取方法简单,适用于工业化生产白桦脂酸。
Objective To establish a method to extract and to determine betulinic acid in semen ziziphus spinosae. Method The betulinic acid was extracted with 95% ethanol, and purified with petroleum ether, 1% sodium hydroxide, and water. Infrared spectrometry (the methanol solution of the extract was scanned) and ultraviolet spectrometry (the extract in methanol was 20 μg·mL -1 ,and was scanned within 190-400 nm) were used as qualitative analysis. HPLC[stationary phase: ALCHROM TM HyperSil C 18 ; mobile phase: methanol:water:triethylamine (92∶8∶0. 04); flow rate: 0. 8 mL·min -1 ; detect wavelength: 210 nm] was used as quantitative analysis. Results By comparing to the standards of UV and IR spectrum, the crystal obtained from semen ziziphus spinosae was betulinic acid. The content of betulinic acid in semen ziziphus spinosae was 0.084%,the purity of the extracted betulinic acid was 86.6%. Conclusion The extraction method is simple and efficient. The quantification determination of betulinic acid by HPLC is accurate.
