异氟烷对大鼠术后学习记忆的影响 被引量:2

摘要 目的观察吸入异氟烷2 h对大鼠趾部切口术后1周和3周空间学习记忆的影响. 方法雄性3个月月龄SD大鼠72只,随机分为6组:对照1周组(Ⅰ组),模型1周组(Ⅱ组),异氟烷1周组(Ⅲ组),对照3周组(Ⅳ组),模型3周组(Ⅴ组)和异氟烷3周组(Ⅵ组),每组12只.除Ⅰ组和Ⅳ组外,其他各组按Brennan法制作趾部手术切口模型,Ⅲ组和Ⅵ组大鼠在手术切口结束后立即吸入1.5%异氟烷2 h.术后1周,Ⅰ组~Ⅲ组大鼠进行Morris水迷宫测试;术后3周,Ⅳ组~Ⅵ组大鼠进行水迷宫测试,每天4次,连续6天.大鼠的潜伏期和游泳距离由Morris水迷宫图像自动监视系统采集和分析完成.结果自测试第1~6天,Ⅲ组大鼠的潜伏期和游泳距离与Ⅰ组和Ⅱ组相比差异无显著性(P>0.05).术后3周,Ⅳ组、Ⅴ组和Ⅵ组大鼠的潜伏期和游泳距离相比差异也无显著性(P>0.05). 结论吸入1.5%异氟烷2 h对大鼠趾部切口术后1周和3周空间学习记忆无明显影响. Objective To investigate the effect of isoflurane inhalation (2 h) on spatial learning and memory in plantar incised rats.Methods Seventy-two three-month-old Sprague-Dawley (SD) male rats weighting 200~ 250 g were randomly divided into 6 groups of 12 animals each: control 1 week group (group Ⅰ), model 1 week group (group Ⅱ), isoflurane 1 week group (group Ⅲ), control 3 weeks group (group Ⅳ), model 3 weeks group (groupⅤ) and isoflurane 3 weeks group (groupⅥ). Except for the group Ⅰand Ⅳ, the rats in group Ⅱ and Ⅲ were made a planter incision according to the methods of Brennan. The rats in group Ⅲ and Ⅵ inhaled 1.5% isoflurane 2 h immediately after planter incision. The Morris water maze testing was performed 4 times every day for 6 consecutive days 1 week or 3 weeks after planter incision. The latency and swimming distances were recorded automatically by the Morris water maze monitoring system.Results From the 1st to 6th day of Morris testing 1 week after planter incision, there was no significantly difference in the latency among group Ⅲ,the group Ⅰ and the group Ⅱ(P> 0.05 );There was no difference in swimming distances among the group Ⅲ, the groupⅠand the group Ⅱ(P> 0.05 ). At 3rn week after planter incision, the latency and swimming distances showed no difference among the three groups(P> 0.05 ).Conclusion 1.5% isoflurane inhalation (2 h) has no effect on spatial learning and memory in plantar incised rats.
