摘要
目的 制作靶向脂质声学造影剂并评估其体外靶向造影成像的可行性。方法 制作高表达αvβ3 的黑色素瘤细胞K173 5的肿瘤细胞模型,利用新桥医院自行研制的脂质造影剂“脂氟显”,通过碳化二亚胺偶合技术,将单克隆抗体与微泡表面物质结合,制作成靶向微泡。分别与高表达αvβ3 的黑色素瘤细胞和低表达αvβ3 的黑色素瘤细胞进行体外结合实验,同时使用水流冲刷实验来验证靶向微泡与靶标结合后的抗流体剪切力的能力,从而在体外条件下研究微泡的靶向特性。结果 建立αvβ3 高表达的肿瘤细胞模型,并成功地制作出靶向肿瘤血管αvβ3 整合素的微泡,加入抗体在45ug以上时靶向抗体结合更加充分,体外试验结果提示靶向微泡与靶标有特异性结合能力,具有一定的抗流体剪切力能力。结论 靶向微泡体外试验结果提示进一步的体内靶向诊断和治疗是可行的。
Objective Microbubbles targeted to v-integrins were prepared and it targeting ability was assessed in cultured cells.Methods To culture K1735 and C23 melanin cell ,then microbubbles targeted to v-integrins were prepared using coavalent means (carboxylate activation with water-soluble carbodiimide) of attachment of the ligand (monoclonal antibody against murine v-integrins) to the performed lipid microbubbles “Zhifuxian” develoed by XinqiaoHospital. Targeted microbubbles combined with K1735 cell of high expression αvβ 3 integrin and C23 cell less expression αvβ 3 integrin, respectively and washed out with water to test the power against flow shear in vitro.Results We constructed successfully K1735 and C23 melanin cell models and prepared targeted lipid microbubbles. A tighter conjugation was set up between the bubble and the target when over 45ug monoclonal antibody was added in shell materials. Targeted microbubbles can especially bind with the target and be against the flow shear to some extent.Conclusion The preparation of microbubbles targeted to αvβ3 integrin was is feasible in vitro.
