长春新碱对K562细胞plk1和γ-微管蛋白表达影响的研究 被引量:12

摘要 目的研究长春新碱对K562细胞周期及plk1和γ微管蛋白影响。方法分析长春新碱处理后K562细胞周期变化,检测长春新碱及plk1反义寡核苷酸处理后plk1andγ微管蛋白表达。结果长春新碱诱导K562细胞G2/M期阻滞。长春新碱影响plk1的聚集和γ微管蛋白形成中心体,plk1反义寡核苷酸同样可影响γ微管蛋白形成中心体。结论长春新碱诱导K562细胞G2/M期阻滞的机制可能是通过抑制plk1的聚集和γ微管蛋白形成中心体来实现的。 Aim To study the change of cell cycle and the expression of plk1 and γ-tubulin in K562 cells treated with vincritine.Methods To analyze cell cyle of K562 cells treated with vincritine, to detect the expression of plk1 and γ-tubulin by western blot and confocal microscopy after K562 cells were treated with vincristine and antisense oligonucleotide of plk1.Results Vincristine induced the arrest of G_2/M phase of K562 cells. Treatment of vincristine influenced the condensation of plk1 and assembly of γ-tubulin. Treatment of antisense oligonucleotide of plk1 also destructed formation of centrosome.Conclusion The function of vincristine might be realized through restraining condensation of plk1 and destruction of centrosome which might be dependent on plk1.
