摘要
目的:观察 TGF β1 对大肠癌细胞增殖与侵袭能力的影响。方法:体外培养大肠癌细胞株 SW480 和LS174T,加入不同浓度的 TGF β1,应用 MTT法检测不同浓度的 TGF β1 对 SW480 和 LS174T细胞增殖的影响,MatrigelTM侵袭试剂盒检测不同浓度的TGF β1 对 SW480 和 LS174T细胞运动和侵袭能力的影响,免疫细胞化学(SP法)检测SW480和LS174T细胞TGF βRⅡ蛋白和Smad 4蛋白的表达。结果:不同浓度的TGF β1(0.1, 1.0,10.0μg·L-1)对SW480和LS174T细胞的增殖均无影响(P>0.05)。TGF β1 能显著促进 SW480 细胞的运动与侵袭能力, 3 种浓度的 TGF β1 (0. 1, 1. 0, 10. 0μg·L-1 )作用于 SW480 细胞 24 h 后,侵袭率分别为 30. 2%、41.1%和41.7%,与对照组的侵袭率18.6%相比明显升高,而在 LS174T细胞中却观察不到此种效应。免疫细胞化学显示 SW480 细胞表达 TGF βRⅡ蛋白,但不表达 Smad 4 蛋白;而 LS174T细胞表达 Smad 4 蛋白却不表达TGF βRⅡ蛋白。结论:TGF β1 能促进SW480细胞的侵袭能力,这种作用不依赖于Smad 4信号途径,TGF β1 发挥生长抑制和促进侵袭能力的作用可能是通过不同的信号路径实现的。
Objective: To investigate the effect of TGF-β_1 on the proliferation and invasion of human colon carcinoma cells in vitro. Methods: Two human colon carcinoma cell lines SW480 and LS174T were cultured in vitro. After exposed to different concentration of TGF-β_1, the proliferation of SW480 and LS174T cells was measured by MTT assay. The expression of Smad4 and TGF-βRⅡ protein of SW480 and LS174T cells were detected by immunocytochemistry. The migration and invasion were determined by BD BioCoat TM Matrigel TM invasion chambers. Results: The proliferation of SW480 and LS174T cells treated 24 hours with TGF-β_1 of different final concentrations (0.1, 1.0, 10.0 μg·L -1 )had no significantly changes compared with that of the controls (P> 0.05 ). The invasive capacity of SW480 cells treated 24 hours with TGF-β_1 of different concentrations(0.1, 1.0 , 10.0 μg·L -1 ) significantly increased compared with that of the controls (P<0.05). The invasive rates of SW480 cells treated 24 hours with TGF-β_1 of concentrations 0.1 μg·L -1 , 1.0 μg·L -1 and 10.0 μg·L -1 were 30.2%, 41.1% and 41.7%, respectively. The invasive rate of SW480 cells treated without TGF-β_1 was 18.6% (P<0.01). The invasion was not observed in LS174T cells. SW480 expressed TGF-βRⅡ protein but did not express Smad4 protein. LS174T cells expressed Smad4 protein but did not express TGF-βRⅡ protein. Conclusion: TGF-β_1 could promote the invasive capability of SW480 independed on Smad4 signaling pathway. The effects of growth inhibition and pro-invasion of TGF-β_1 might be carried out via different signaling pathways.
