摘要  目的 观察重组人肿瘤坏死因子(rhTNF)- α和5- 氟尿嘧啶(5 FU)对人胃癌细胞株的生长抑制作用及其作用前、后人胃癌细胞株中死亡相关蛋白(DAP)3 的表达情况,探讨 DAP3 的临床意义。方法 选择不同浓度的 rhTNF α(0、50、100、200 ng/ml)和5 FU(0、1×10-6、1×10-5、1×10-4 mmol/L)作用于人胃癌细胞株BGC 823和HGC 27,采用酸性磷酸酶法测定各组细胞株的生长情况,确定rhTNF α和5 FU的有效浓度。采用蛋白质印迹法测定经有效浓度的rhTNF α和5 FU处理前、后人胃癌细胞株BGC 823和HGC 27中DAP3的表达情况。结果 酸性磷酸酶法测得100 ng/ml的 rhTNF α和1×10-5 mmol/L的5 FU可有效地抑制人胃癌细胞株 BGC 823 和 HGC 27 生长。人胃癌细胞株 BGC 823 和 HGC 27 中均未见 DAP3 表达,经 100 ng/ml 的rhTNF α或1×10-5 mmol/L的5 FU处理后 48 h,人胃癌细胞株 BGC 823 和 HGC 27 中均可见 DAP3 表达。结论 rhTNF α和5 FU通过诱导DAP3表达可抑制人胃癌细胞株BGC 823和HGC 27的生长,推测 DAP3 为一新的肿瘤治疗位点。 Objective To investigate the expression of death-associated protein 3(DAP3) in two human gastric cancer cell lines: BGC-823 and HGC-27 as well as its change after treatment with rh-tumor necrosis factor(TNF)-α and 5-FU.Methods Two human gastric cancer cell lines: BGC-823 and HGC-27 were treated with rhTNF-α and 5-FU at different concentrations and the treatment concentration was determined by acid phosphatase method. The expression of DAP3 before and after treatment with rhTNF-α and 5-FU were determined by Western blot method. Results The treatment concentrations of rhTNF-α and 5-FU were 100 ng/ml and 1×10 -5mmol/L respectively. The DAP3 could not be detected in both cell lines before treatment but could be detected after the treatment.Conclusions RhTNF-α and 5-FU inhibit human gastric cancer cell lines via inducing expression of DAP3 which may be a potential target for gastric cancer treatment.
