摘要
应用多聚酶链反应(PCR)和脱氧核糖核酸(DNA)重组技术,构建了人白细胞介素6(IL-6)重组表达载体pTSMIL-6。利用DEAE-Dextron法将该表达载体转染COS7细胞,获得了有生物活性IL-6的暂时性高效表达。对收集的上清进行检测表明:IL-6的生物活性在转染后24小时为14208IU/ml,48小时为4770IU/ml;SDS-聚丙稀酰胺电泳证明有IL-6糖蛋白区带,分子量为25794.6D(26kD)。
Using PCR and DNA recombination methods, we have constructed a human rIL-6 expressing clone pTSMIL-6.
Using DEAE-Dextron method, the COS7 cells were transfected by pTSMIL-6. The transfected cells expressed high level of IL-6, but the expression showed transiently. The amount of IL-6 in collective supernatant was 14 208 IU/ml in 24h after transfection, and 4 770 IU/ml in 48h. COS7 cells did not secrete IL-6 spontaneously. SDS-PAGE electrophoresis showed a MW 26kD protein band of IL-6.
