摘要
本文建立了一个能灵敏和精确地测定脑组织中低水平的RnE2,并能观察到生殖功能衰退时老年大鼠RnE2量变化的方法。对雌性大鼠垂体前叶(AP)及大脑三个脑区:视前区(POA)、丘脑下部内侧基底部(MBH)和杏仁核(AMG)的雌二醇核受体(RnE2)进行了测定,结果显示:1.中年大鼠AP、POA、MBH和AMG中RnE2的含量以AP中最高,AMG中最低;2.青年和中年大鼠的AP和三个脑区中RnE2的含量是动情前期比间情期高出一倍以上;3.老年大鼠AP中RnE2的含量比青年和中年两个组低;4.经植入孕酮或摘除一侧卵巢后的老年大鼠,在AP和三个脑区中RnE2的量与正常老年大鼠相比出现波动。
A sensitive and accurated method for the assay of RnE2 was established and proved to be suitable to determin the low levels of the RnE2 in the brain tissues of rats and to investigate the effect of the aging reproductive function on the changes of RnE2 level. The nuclear estradiol receptor (RuE2) contents in the anterior pituitary (AP) and the three brain areas, ie. preoptic area (POA),medial hasal hypothalamus (MllH) and amygdala (AMG) of the female rat were determined. The results showed: 1. In the AP, POA, MBH and AMG of the middle aged rats, the RnE2 levels in the AP were the highest and lowest in the AMG; 2. In the AP and three brain areas of the young and middle-aged rats, the RnE2 levels at proestrus in both groups were one-fold higher than those at diestrus; 3. The RnE2 levels in the AP of the aging female rats were lower than those of young and middle-aged groups; 4. After progestin implantion or hemi-ovarectomy of the aging female rats,the RuE, levels in the AP and the three brain areas fluctuated as compared with those of the normal rats.
