摘要 本文采用无乳化剂乳液聚合法,以苯乙烯、丙烯酰胺、丙烯酸及有机染料为原料,合成了大粒径彩色高分子微球,并通过两步化学反应在微球表面衍生出可与蛋白质迅速反应的叠氮羰基。探讨了聚合条件对微球粒径大小的影响,并测定了微球的红外光谱及结合蛋白量。实验结果表明:在偏酸性条件下加入适量有机溶剂、电解质及油溶性染料可制得大粒径彩色高分子微球。微球的红外光谱图在2140cm^(-1)处有吸收峰,表明微球表面有叠氮羰基。电镜法测得微球的平均粒径为2.50±0.04μm。微球结合牛血清白蛋白的量为3.2mm·m^(-2)。 The colored microspheres of styrene/acrylamide/acrylic acid/dye (St-AAm-AA-Dye) copoly- mer with azidocarbonyl groups on the surface were prepared in the following steps: 1. Synthesis of St-AAm ?- AA-Dye microspheres by means of emulsifier-free emulsion polymerization; 2. Hydrazinolysis of surface acrylamide groups; 3. Preparation of azidocarbonyl groups on the surface by treating the hydrazide micro-spheres with sodium nitrite and HCI. The effect of the polymerization conditions on microsphere'size was investigated. The microspheres were characterized by IR and transmission electronic microscope. The result of our experiment indicated that monodisperse and colored large polymer microspheres having diameter 2. 5±0. 04 μm were obtained by addition of certain organic solvent, electrolyte and dye at pH 4. 5-5. 0. The azidocarbonyl groups on the microsphere surface can covalently combine with the free amino groups of proteins under bland conditions through peptide bond. These big and colored microspheres can be observed under an optical microscope and can be used for labeling cell-surface antigens.
