衰老过程中鼠肝细胞核体外重组的转录活性 被引量:2

Transcriptional Activity of Reconstituted Nuclei of Rat Liver in Aging Process

摘要 本文以0.35mol/LKCl抽提不同年龄鼠肝细胞,将抽提后细胞核分别与抽提物、纯化的染色质高迁移率非组蛋白(HMG)及组蛋白H1进行重组。结果发现,0.35mol/LKCl抽提后老年鼠肝细胞与断乳鼠肝细胞核抽提物重组转录活性高于其与自身或青年鼠肝细胞核抽提液重组者。还发现高迁移率非组蛋白可提高抽提后鼠肝细胞核转录活性,对断乳鼠的作用最强;但不影响未经抽提的细胞核转录活性。相反,组蛋白H1则可抑制各年龄组鼠肝细胞核的转录活性。 Nonhistone protein in rat liver nuclei was extracted by 0.35 mol/L KCl. The extracted nuclei were reconstituted with the extract of rat liver nuclei, non-histone high mobility group protein of chromatin(HMG) and histone H1 respectively. Transcriptional activity of the old rat liver extracted nuclei reconstituted with extract of weanning rat liver extracted nuclei ,was higher than reconstituted with extract of old or young one. In addition,while HMG was used in stimulating the transcriptional activity of the extracted nuclei the stimulation effect on the weanning was stronger than on other one,but HMG had no effect on unextracted nuclei. Contrast to HMG histone H1 showed obvious inhibition effect on transcriptional activity of all of the rat liver nuclei.
