大黄抑制糖尿病鼠肾肥大和肾小球多肽生长因子表达的关系 被引量:5

摘要 本文观察了中药大黄醇提物对糖尿病大鼠肾小球内多肽长生因子的影响。结果表明,糖尿病初期虽还未见肾重量的明显增加,但肾小球内生长用于的表达已显著“上调”,以链脲菌素注射后24h最明显,大黄治疗后,其多肽生长因子表达显著“下调”,并且在治疗96h后肾脏重量亦明显低于非治疗组。因此,说明大黄抑制糖尿病大鼠肾脏肥大的作用与其“下调”肾小球内多肽生长因子的表达有关。 e studied the glomerular mRNA levelsenceding for propre EGF, PDGF-b chain , andTGF in SD rats 12, 24, 48 and 96 hours afterinjection of Streptozotocin. At 24 houre mRNAlevels measured by dot hybridization for propreEGF,PDGF-b chain, and TGFβ1 increased 2. 8-fold (P< 0. 05) , 3. 1-fold(P<0.01) ,and 7.3-fold (P<0. 01 ) , respectively ; at 48 hours mRNAlevels for these growth factors peaked in theglomeruli of diabetic rats as compared withcontrol rats. RO treatment partially amelioratedthe increase of mRNA levels for propre EGF,PDGF-b chain and TGFβ in the glomeruli ofdiabetic rats. Renal hypertrophy was obeerved at72 hours in the diabetic rats ,but not in the RO-treated rats. We concluded the abnormalproduction of growth factors in the glomeruliwas involved in the early renal hypertrophy indiabetic mellitus and the effects of RO on thesuppression of the renal enlarrgement in diabeticrats may be related to its effect of down-regulated sene expression of growth factors inthe glomeruli.
