摘要
研究了以质粒为模板Sanger双脱氧链终止法测定DNA序列的操作条件。用本实验改良的制备质粒模板方法和测序反应程序,使以32P为DNA合成标记、双链质粒为模板的测序结果基本达到以35S以33P为标记、M13单链DNA为模板的测序效果,实验操作简便、成本低。
Procedure involved in DNA sequencing using a plasmid as a template anddomestic isotope (32) ̄P as a labelling precursor based on Sanger's dideoxy-chain-terminating method was optimized.The sequencing autoradiograph resulted from the improvedmethod proved to be almost the same in resolution,easier in operation and much less incost compared with the method using M13 as a template and(35) ̄S or (33) ̄P as a labelling precursor.
