摘要
研究了以质粒为模板Ｓａｎｇｅｒ双脱氧链终止法测定ＤＮＡ序列的操作条件。用本实验改良的制备质粒模板方法和测序反应程序，使以３２Ｐ为ＤＮＡ合成标记、双链质粒为模板的测序结果基本达到以３５Ｓ以３３Ｐ为标记、Ｍ１３单链ＤＮＡ为模板的测序效果，实验操作简便、成本低。
Procedure involved in DNA sequencing using a plasmid as a template anddomestic isotope (32)￣P as a labelling precursor based on Sanger's dideoxy-chain-terminating method was optimized.The sequencing autoradiograph resulted from the improvedmethod proved to be almost the same in resolution,easier in operation and much less incost compared with the method using M13 as a template and(35)￣S or (33)￣P as a labelling precursor.
