摘要
从收集的5株枯草芽孢杆菌(Bacillussubtilis)菌株中,筛选出一株具有一定产肌苷能力(10g/L),并带腺嘌呤缺陷型遗传标记的菌株.以该菌株为出发株,通过60Coγ-射线和亚硝基胍复合诱变,得到一株对8-氮杂鸟嘌呤具有抗性,并且丧失了部分核苷磷酸化酶活性的突变株.在此基础上,通过正交试验,得到肌苷发酵最佳条件,在5L全自动发酵罐上进行了多次试验,肌苷产量达到15g/L.
The mutant HW-9 of Bacillus subtilis showing the property of Ade-,NP-,8-AGr, was selected from five strains by the methods of 60Co γ-ray and chemical NTG mutation.The optimal fermentation technology was got by orthgonal design.After four times of fermentation on five liter fermentor,the concentration of inosine in fermentation broth was about 15g/L,which was 31% more than that of original strain.
