摘要
神经内分泌系统与免疫系统间存在着双向联系,它们通过共有的一套激素、细胞因子等分子递质相互调节。一些文献表明:TNFα与hCG似参与共同的病理生理过程。我们对TNFα与hCG在体外诱生时可否相互调节进行了初步探讨。即hCG对LPS激活的单核巨噬细胞产生TNFα的影响.并用抗hCG-β单抗阻断试验证明hCG作用的特异性。另一方面,据文献报道及我们的有关工作。混合淋巴细胞培养上清中有irhCG的产生,我们在此诱生体系中,加入rTNFu或抗rTNFα单抗阻断后的rTNFα,以观察irhCG产生是否受到影响。结果发现,TNFα与hCG在体外诱生时可相互抑制。这种相互抑制作用在母胎关系、移植排斥及肿瘤等疾病中可能有一定的病理、生理意义。
The neuro-endocrine
system and im mune system interregu tate through a series of shared
messengermolecules,such as hormones and cytokines.Some publication suggested that TNFα
and hCG seem to be involved in some scribed the interaction of the two agents during their
production in vitro.The production of lymphocyte-derived hCG or irhCG,in mixed lymphocyte
culture(MLC)has been reported by our previous work and reports from others.In this study rTNF
α was added to the MLC system(33MLCsamples in all).the condensed supernatants were
detected for irhCGβby RIA.The results showed that irhCG production in MLC was greatly
inhibited by the addition of rTNFα On the other hand,hCG or anti-hCGβMcAb wer added
respectively to the LPS-stimulated M cultures to observe the production of TNFαThe results
showed that hCG had a suppressive effect on TNF produced M and this suppres-sion was
abolished only by anti-hCGβ McAb but not irrelevant McAb.It suggested that the suppressive
effect of hCGH onTNFαproduction was at least related to the β-subunit of hCG.Taken together,
our study demonstrated that the two agents may down-regulate each other.By reviewing related
articles,We postulate that some suppressor mediators may be in-duced,such as PGE.Our
results also in dicated that the interaction of cytokines and homones may be involved in the
pathological mechanism of some diseases.
