摘要
本文应用兔抗ｈＡＦＰＩｇＧ包被板条，以异硫氰酸苯基－ＥＤＴＡ·Ｅｕ￣３＋标记抗ｈＡＦＰＭｃＡｂ，建立了固相“夹心”二步法分析ｈＡＦＰ技术。结果显示。分析范围是６．２５ｎｇ／ｍｌ～１００ｎｇ／ｍｌ（ｒ＝０．９９），变异系数在１．６７％～１７．７４％之间，ｈＡＦＰ最小可测值为０．３５ｎｇ／ｍｌ．测定７６例孕妇血清样品，同所购ＤＥＬＦＩＡｈＡＦＰ药盒结果的相关性（ｒ＝０．８８）好。该法检测人血清ｈＡＦＰ的特点是操作方便、成本低。
ln this Rh-IFMA of hAFP,strip-wells were anted with a polyclonal
antithey against hAFP.To prepare the lable,a monoclOnal antihbody was derivatized with
isothiocyanatophenyl-EDTA-Eu￣(3+). In the solid-phase sandwich as-say,a twr-step procedure
was used;the measuring range was 6. 25ng/ml～100 ng/ml(r=0. 99).The CV within the as-say
range was between 1. 67 and 17.74%,depending on the dase.The minimum detectable
coneentration of hAFP was 0. 35ng/ml. A total of 76 samples from maternal serum were tested.
Correlation of the results between this method and DELFIA kit was good(r=0. 88 ).Our methed for
measuring hAFP in human serum is both convenient and inexpensive.
