摘要
本实验应用杂交瘤技术,首次在国内建立了两株抗人补体终未阶段同源限制因子(HRF)的杂交瘤细胞系(2A7和1F10)。经抗原特异性及功能分析提示它们所针对的抗体分子具有相似的功能,但抗原性和分子量不同:2A7-Ag为18~22KDa,1F10-Ag为65KDa。进一步应用免疫荧光流式细胞分析、促同源补体溶血试验、中和实验、Westernblotting、PI-PLC处理等实验手段系统地鉴定了2A7抗原的细胞分布、抗原特异性、分子特性及功能。表明2A7与CD59的命名抗体MEM43和1F5共同识别同一膜糖蛋白分子,该分子以糖脂链(GPI)锚固于细胞膜上,证明2A7为一株抗CD59的单克隆抗体。
Two hybridomas.clone 2A7 and 1F10 were obtained by screening fOr antibcaly which caused hemolysisof human erythrocyte by human and EDTA-HuS. They react with two different proteins with MW of 18~22KDaand 65KDa respectively. Flow cytometric analysis showed that mAb 2A7 recognizes an antigen expressed on all peripher-al leucocyte and several cell lines.but was absent from U937 and Raji cell lines. This antigen was sensitive to treatmentwith pheephdylinosito-specific pheepholipase C(PI-PLC) on Jurkat cell line but not on erythtocyte. Compare mAb 2A7witb two known CD59 antibodies MEM4 3 and 1F5.it was found that three mAbe all recognized related epitopo on thesame erythrocyte glycoprotein. These results demostrated that 2A7 is a new monoclonal antithey against CD59.
