摘要
本研究建立了适用于快速检测空肠弯曲菌的吖啶橙免疫荧光苗团培养沉淀法。用该法可检出每毫升接种约40个菌,增菌24h的培养物或每毫升合约10万个菌的菌悬液,不与水弧菌、大肠杆菌等13种杂菌发生交叉反应。对貉、水貂、獭兔和熊等经济动物310份肛拭样本进行了检测,其阳性率分别为66%、19%、21%、29%,与常规培养法均相差不显苦(P>0.05),可在27h内报告结果,而常规培养法则需要5~7d。该法适用于大样本的检测和基层单位应用。
An acridine orange immuno-fluorescent bacterial clumpmethod for detecting Campylobacter jejuni in economical animals hasbeen established. In preliminary expriment the positive results could beobtained from bacterial suspension of 1.0×10~5 cells/mL or 24 hoursculture in selective enriched broth which had been inoculated with 40cells/mL. No cross reaction took place with 13 species of bacteria otherthan Campylobacter jejuni. The positive rates of 310 fecal samplesfrom racoon dog (125), mink (103), rex rabbit (68) and bear(14) were66%, 19%, 21% and 29%, respectively. Compared with the routinecultivation method, there was no significant difference (P>0.05)between them. However, the routine culture method takes more time thanimmuno-fluorescent bacterial clump method.
