摘要
以美味猕猴桃东山峰78-16的茎段愈伤组织块为受体,根癌农杆茵(Agrobacteriumtumefaciens)LBA4404(pBI121)作介导菌,将愈伤组织块与农杆菌共培,扫描电镜观察农杆菌在愈伤组织块细胞表面上的附着情况;将共培后的愈伤组织块转移到含50μg/mL的卡那霉素(Kan)筛选培养基上筛选,获得抗Kan的抗性苗。抗性苗的获得初步证明外源新霉素磷酸转移酶Ⅱ(NPTⅡ)基因已整合到受体基因组中并得以表达。
Transform the steam-derived callus of kiwi Actinidia deliciosa(A. Chev)C. F. Liang et A.R. Ferguson with A. tumefaciens LBA4404(pBI12l).After co-cultural ex- periment,the circumstance of A. tumefaciens attaching to the surface of callus was observed with scanning electron microscope and the co-cultural materials were transfered onto the screening cultures medium containing 50 μg/mL kanamycin. In the result of kanamycin-resis- tant screening, four resistant plantlets have been obtained,which demonstrates that exoge- nous NPT Ⅱ gene has expressed in acceptor.
