急性重度低压缺氧对大鼠心肌脂质过氧化与心肌酶活性的影响 被引量:15

Investigation of Myocardial Lipid Peroxidation and Myocardialdial Enzyme Activity under Acute Hypobaric Hypoxia

摘要 为了观察急性重度低压缺氧对心肌脂质过氧化及心肌酶学的影响,对39只成年健康Wistar大鼠急性低压缺氧实验。结果发现,急性缺氧后心肌组织中硒谷胱甘肽过氧化物酶(SeGSHpx)活性明显降低,丙二醛(MDA)含量减少,SeGSHpx/MDA比值下降。结果提示,急性低压缺氧后,心肌组织中的脂质过氧化反应并无明显增强,仅有心肌的潜在抗氧化能力降低。另外,急性低压缺氧后,心肌酶活力显著增高,说明急性低压缺氧对心肌有一定损伤作用。鉴于心肌脂质过氧化反应无明显增强,提示心肌损伤与脂质过氧化无明显相关。心肌酶活力增高的原因,可能是急性缺氧对心肌细胞直接作用的结果。 The damage caused by acute hypobaric hypoxia upon myocardium is apotential danger for pilots flying in high altitude. With the advance of the investigation ofmyocardial ischemia-reperfusion injury , it was found recently that the increase of oxygenfree radical production and the strengthening of lipid peroxidation reaction were importantcauses for the development of ischemic myocardial damage.The development and the progress of atherosclerosis were also closely related to the damage from lipid peroxidation. In this article , SeGSHpx , MDA content , SeGSHpx/MDA ratio and several kinds of my-ocardial enzymes such as CPK,CK-MB,LDH,α-HBD and AST were measured in 39 healthy adult Wistar rats staying at 7500m for 30min in a hypobaric chamber in order toobserve the effect of acute hypoxia upon the myocardium and the relationship between lipid peroxidation and myocardial dalnage. The results showed that activity of myocardialenzymes was raised significantly after acute hypoxia,the drference was remarkable ascompared with the control values(P<0.01 or 0.00 1). Acute hypobaric hypoxia had al-so some infiuences on the myocardial SeGSHpx activity , MDA content , and SeGSHpx/MDA ratio. Normal organism has the ability to combat peroxidation with sufficient oxy-gen supply , the human body can generally tolerate the influence of oxygen free radicalsupon myocardiai tissue by the dynamic balance of the production of intracellular peroxi-dates and the protective mechanism of the antioxidant system. The change of the activity of SeGSHpx in rats in the hypoxic group were much lower than that in the control group,the difference was very significant(P<0.001) which indicates the inefficiency of my-ocardium against peroxidation. MDA is the end product of lipid peroxidation , and the ele-vation of plasma MDA content reflects that the lipid peroxidation of the body is exacer-bated. In this article , the myocardial content of MDA of rats in the hypoxic group waseven lower than that in the control group , the difference between the two groups was sig-nificant(P<0.05). The results suggested that there was no exacerbation of lipid peroxi-dation after severe hypoxia.SeGSHpx / MDA ratio reflects the combined change of the above two factors. Myocardial SeGSHpx / MDA ratio of rats in the hypoxic group was lower than that in the control group , and the difference was very significant(P<0.01), suggesting the lowering of potential ability of myocardium to combat peroxidation.To sum up , the authors considered that there was no intimate relationship between myocar-dial damage and lipid peroxidation. The cause for the elevation of the activity of myocar-dial enzymes might be the direct result of acute hypoxia upon myocardial cells。
