摘要
高纯度人绒毛膜促佐腺激素(HCG)的制备方法已有许多报道,通常都采用DEAE-纤维素离子交换层析法和磊胶过滤法反复槽制,价格昂贵.我们采用价格低廉的磷酸纤维赐作为寓子交换剂,宋代沓DEAE-纤维素,对具有历-20iu/mp生物效价的HCG粗品进行纯化,纯化后的HCG溶液,其紫外扫描图谱(220—300urn)与典型的蛋白质扫措图谱一致J经聚丙烯醉胺凝胶电泳分析,可得一条清楚的蛋白质条带;SDS-聚丙烯酞胺凝胶电泳分析,可见两条清晰的蛋白质色带.
Many methods have been reported for high purification of HCG. but those are usually purified with expensive DEAE-Cellulose. This paper presents a method to purify HCG using cheap cellulose phospho-rylated instead of DEAE-Cellulose. The chromatogram(220-300nm) of purified HCG is in conformity with the typical chromatogram of protein. The results show that HCG purified has one band in poly-acryla mide gel electrophoresis(PAGE) and two bands in SDS-PAGE.
