血清吗啡放射免疫分析

摘要 用混合酸酐法合成的吗啡-6-琥珀酸-BSA作为免疫原,制备了高特异性和高亲和力的抗吗啡血清.用氯胺T-法制备^(125)l-吗啡,建立了血清吗啡放射免疫分析方法.并已应用于临床,获得了满意的结果.本法灵敏度为1ng/ml,平均变异系数,批内4.06%、批间8.25%,正常值9.01±1.74ng/ml,无性别差别.本法对27例鸦片类成瘾者检测,其血清吗啡浓度均高于正常值. Morphine is an active metabolite of opiate analogueg.The radioimmunoassay of morphine in serum was established. The morphine-6-succinyl-bovine serum albumin was immunizator was synthesized. Thespecific antiserum was obtained from the immunizing rabbits. The 125I-Morphine was prepared by thechloramine-T method. The result of methodological studty showed that the sensitivity was 1 ng/ml ,theaverage recovery was 106% ,the intra-and inter- C. V. was 4.06% and 8.25% respectively. The normalserum concentration of morphine was 9.01 ±1.74 ng/ml. No significant difference was observed in different sex groups. The serum concentration of morphine or opiate analogues in addict was much higherthan that in normal. This new method can be used as an effective tool to judge the degree of addiction to the opiate analogues and plays a role in the administration.
