摘要
采用正常Wistar大鼠,雌雄不拘。老龄鼠(20个月)8只,壮龄鼠(8个月)13只。采用石蜡切片,ABC法染色,DAB显色。光镜下观察下丘脑ACTH-ir和β-END-ir阳性细胞形态、分布和计数。图像分析仪测其灰度值。结果:弓状核β-END-ir阳性细胞可分为两种类型,Ⅰ型为胞核不明显,着色较深,数量较多的强阳性细胞;Ⅱ型为胞核明显,着色较淡,数量较少的弱阳性细胞。ACTH-ir阳性细胞,胞核明显,胞体较大,类似β-END-ir弱阳性细胞。老龄大鼠两种阳性细胞出现率均明显低于壮龄大鼠(P<0.05)。老龄大鼠弓状核ACTH-ir阳性细胞灰度值显著低于壮龄大鼠(P<0.01),其β-END-ir阳性细胞灰度值则显著高于壮龄大鼠(P<0.01)。并对结果进行了讨论。
In this study, the normal female and male Wistar rats.8 aged rats(twenty-month-old)and 13 adult rats(eight-month-old) were used.The blocks containing hypothalamus were dehydrated and embedded in paraffin(50-52℃)The sections(7μm) were stained with ABC technology and coloured by DAB,The morphologydistribution and counting of adrenocotropin-immunoreactive (ACTH-ir)and beta-endorphin-immunoreactive(β-END-ir)neurons in the rat hypothalamus were observed and measured under microscope.The light values of positive neurons were measured with Image Analyser.The results show that β-END-ir neurons may be divided into two types:(1)deep colour positive neurons,which are unclear nucleus or deep stain in colour and numerous and (2) light colour positive neurons,which are clear nucleus or light stain in colour and rarely seen,The ACTH-ir neurons are similar to the second type of β-END-ir neurons,whose nucleus are well defined and the bodies are larger than others,The appearance rated of ACTH-ir and β-END-ir neurons in the aged rats are lower than those in the adult rats(p<0.05).The light value of ACTH-ir neurons in the aged rat arcuate nucleus is obviously lower that in the adult rat arcuate nucleus (P< 0.0001),but that of β-END-ir neurons is opposite(P< 0.0001).
