放射损伤、烧伤和放烧复合伤心肌线粒体脂质过氧化的加强效应

Studies of intensification of lipid peroxidation of myocardial mitochondria after combined radiation─burn injury in rats

摘要 Wistar大鼠经6Gy60Coγ线和/或30%总体表面积(TBSA)Ⅲ度烧伤致放射损伤(RI)、烧伤(BI)和放烧复合伤(RBI).观测伤后1、3、8、16和24h大鼠心肌线粒体脂质过氧化和抗氧化酶活力的改变,发现三种伤类均表现为脂质过氧化代谢产物丙二醛(MDA)含量伤后1h即明显升高,24h降至对照组(C)水平;超氧化物歧化酶(SOD)活力在整个实验过程中维持在明显低于C组的水平;BI组和RBI组还可见谷胱甘肽过氧化物酶(GSH-Px)活力在伤后16和24h明显降低。结果表明,伤后心肌线粒体抗氧化酶活力降低,脂质过氧化加强,提示线粒体内有自由基堆积。结合此前同样改变条件下所见的心功能下降,认为自由基的有害作用可能是心功能降低的重要原因之一。 The content of malondialdehvde(MDA)and the activity of antioxidization enzyrnes of the myocardial mitochondria were observed in rats,which were randomized into the control(C),burn injury(BI),radiation injury(RI)and combined radiation-burn injury(CRBI)groups,in the lst,3rd,8th,16th and 24th hour after injury,The findings were as follows;1,The MDA content was increased significantly in the lst hour and returned gradually to the control level in the 24th hour,The activity of superoxide dismutase(SOD)was kept in an obviously lower level than that of the control throughout the observation period in BI,RI and CRBI groups.2.The activity of GSH-Px was decreased in BI and CRB groups.The fidings indicate that the capacity to scavenge free radicals is weakened and lipid peroxidation intensified in the,myocardial mitochondria of the injured rats,Consequently,it is deduced that the harmful effects of free radicals might be one of the factors to induce cardiac dysfunction,injury under the same injurious in our previous report.
