老化红细胞的分离和鉴定

摘要 根据本实验室的工作,老化红细胞表面可结合自身IgG的特点,建立了一种新的分离年轻及老化红细胞的方法。由于Protein A可与IgG有特异结合,用活化的Sepharose 6MB结合Protein A制成亲和层析柱,以此分离老化红细胞。同时对分离的红细胞作了谷胱甘肽还原酶、谷胱甘肽过氧化物酶、过氧化氨酶、6-磷酸脱氢酶的活性和变形性及吞噬情况的比较。 A new method employing the affinity chramatograpy on a protein A-conju-gated Sepharose 6MB column has been established to isolate human senescent red cells based on the fact that human aged red cells have autologous IgG bound to their surface. The activities of glutathione reductase, glutathione peroxi-dase catalase and G-6PD, in addition to deformability and the speed of being phagocytocized by macrophages of young and senescent red cells separated by this method were compared. The results were shown that the activities of enzymes and deformability of aged red cells were all reduced, erythrocytes affinity chromatograpy senesence
