高效液相色谱法测定血浆及尿中氯霉素的含量

DETERMINATION OF CHLORAMPHENICOL IN PLASMA AND URINE BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

摘要 采用高效液相色谱法,以非那西汀为内标物在λ_(max)=278nm下,研究了血浆及尿中氯霉素的含量。本法测定结果:保留时间:氯霉素7 min,非那西汀11min;最低检测浓度:血样为52.3%μg/ml,尿样为37μg/ml;平均回收率:血样为100.5%,尿样为99.7%;线性范围:血样及尿样均为5~50μg/ml;日内变异系数:血样在1.10%以下,尿样在1.19%以下;日间变异系数:血样在5.3%以下,尿样在5.9%以下;新诺明等药物对测定无干扰。 A selective and sensitive method for the determination of chloramphenicol inplasma and urine by high performance liquid chromatography has been developedwith Phenacetin as internal standard. A ODS column (particle size 10μm, 150×4mm i.d.) was used. The flow rate of mobile phase (a 30% methanol solution)was 1.5ml/min. Ultraviolet detection was made at 278nm. The retention time was7min.for chloramphenicol and 11min. for phenacetin. The limit for detection wasto 52.3μg/ml for plasma and 37μg/ml for urine. The recovery of added chlo-ramphenicol was 100. 5% for plasma and 99. 7% for urine. A calibration curve waslinear in the range from 5μg/ml to 50μg/ml for both plasma and urine.The within-run reproducibilities were 0.92 and 1. 10% (n=10)for plasma and 0.90 and 1. 19%(n=10) for urine at 20 and 40μg/ml. The day-to-day wariabilities were 5.30 and2.56%(n=7) for plasma and 5.90 and 3.80%(n=7) for urine at 10 and 50μg/ml.Possible interference from other drugs is discussed.
