摘要
本实验制备了一种单克隆抗体５Ｈ１２，该单抗不但能与正常人外周血多种白细胞反应，也能与多种细胞株反应，说明５Ｈ１２单抗识别的抗原（即５Ｈ１２）是一种白细胞共同抗原。功能实验表明，５Ｈ１２单抗能够抑制Ａｎｔｉ─μ诱导的Ｂ细胞增殖，提示５Ｈ１２抗原对Ｂ细胞活化有某种调节作用，证明５Ｈ１２抗原是一种分子量为２２ＫＤ的单链蛋白质分子。
We have developed a monoclonal antibody 5H12 that has a wide reactivity with a panel of human leukocytes and white cell lines.The data suggest that the antigen recognized by the monoclonal antibody 5H12(5H12 antigon)is a leukocyte common antigen.Function study showed that monoclonal antibody 5H12 could inhibit the B cell proliferation induced by anti-u in a dose-depcndent manner(1:1000 to 1:62.5).SDS-polyacrylamide gel electrophoresis and Weatern-blotting analysis indicated that 5H12 antigen is a 22KD one-peptide chain protein.
