摘要
本文描述简化微孔沪膜硷洗脱法,并提出药物对肿瘤细胞DNA破裂选择性指数(Is)作为评价及筛选此类抗癌药物的指标。
Some important antitumor drugs induced cellular DNA breaks as theirprimary actions, Comparing the abilities of DNA cleavage in cancer cells with that innormal cells may reflect their antitumor effect. Alkaline elution method was alreadyused to detect the amount of cellular DNA breaks。 In this experiment, alkaline elutionmethod was modified. Instead of scintillation counter, ultraviolet spectrophotometer wasused to monitor the fraction of DNA. Buffer of 0. 05M NaCl-NaOH pH 12.25 was usedas alkaline elution solution in place of tetrapropylaminium hydroxide- EDTA.We propose the Index of Selectivity(Is) of a drug to tumor celI DNA breaks as anindicator for estimating and screening some kinds of antitumor drugs. The formula is asfollows. An logno/n Where at is coefficient of cancer cell DNA breaks: An is coefficient of normal cellDNA breaks; to and t are retaining rates of DNA of cancer cell treated without andwith drug on filter respectively ; no and n are retaining rates of DNA of normal celltreated without and with drug on filter respectively. The is can be detected with cancercells and normal cells under the following conditions: 1. the same concentration of thedrug; 2. the same eluting rate; 3. other experimental conditions were the same.
