重组人肿瘤坏死因子及其与阿霉素联合抗人肝癌的实验研究 被引量:5

EFFECT OF RH-TNE ALONE AND IN COMBINATION WITH ADRIAMYCIN ON HUMAN HEPATIC CARCINOMA

摘要 本研究采用体外细胞毒方法(MTT)检测了重组人肿瘤坏死因子(rHTNF)和(或)阿霉素(ADM)对肝癌细胞株SMMC─7721及Bel─7402的细胞毒效应。结果表明:rHTNF、ADM对两肝癌细胞株的毒性有明显差异,rHTNF与ADM联合应用的体外细胞毒试验显示良好的协同增强效应。在此基础上,选用SMMC─7721细胞株进行棵鼠移植瘤试验,观察rHTNF与ADM联合应用的体内抗瘤效应。在治疗6个周期后,空白对照的肿瘤大小为:2.387±0.97cm3,ADM与rHTNF联合应用组仅为0.43±0.189cm3,两者相比P\0.001。联合用药对肿瘤生长的抑制率为82.07%,明显高于单用ADM组(38.83%)和单用rHTNF组(27.3%),联合用药的协同增强抑制率为15.94%。由此再次证明:rHTNF与ADM联合抗瘤作用并非简单的作用叠加,体外及裸鼠体内试验均显示协同增强的抗瘤效应。本实验为肝癌的治疗提供了实验依据。 In order to investigate the value of tumor necrosis factor(TNF)combined with adriamycin against human hepatocarcinoma, we have used MTT method to examine the in vitro cytotoxic activity of rhTNF or and ADM against two human hepatocarcinoma cell lines: SMC-7721 and BEL-7402. The results showed that the cytoto-xic effects of rhTNF against these two cell lines were different. We found that rhTNF singnificantly enhanced the cytotoxic activity of ADM. Based on experiments in vitro, we have examined also in vivo tests. using SMMC-7721 tumor-bearing nude mice. The anti-tumor effect was evident. The addition of rhTNF to ADM resulted in a singnificant re-duction in tumor volume at six cycle treatment compared to untreated animals. The ave-rage volumes of the tumors were 2.387±0.97cm3 and 0.43±0.189cm3 respectively (p<0.01). The tumor growth inhibition rate was 82.07 % for the combination group as compared to that of 38.83% for ADM alone group and 27.3% for the rhTNF alone group. These result showed that rhTNF plus ADM had a synergistic enhancement of antineoplastic activity in vivo. In conclusion, from the study of vivo and vitro, we believe that some hepaticcarcinoma patients will be benefit from receiving treatment with rhTNF plus chemotherapy.
