摘要
外源性凝集素作为肿瘤碳水化合物抗原研究的理想工具已引起人们的重视。本研究用外源性凝集素对4株胃癌单克隆抗体相应抗原进行了分离、鉴定和检测。首先用刀豆凝集素亲和层析分离胃癌组织N-糖苷型糖蛋白,并用单抗检测证实单抗Md1,MG7、MGb1、MG1相应抗原均能被刀豆凝集素识别,其中MGdl、MG7也可被PHA-E所识别Lectin-MG夹心ELlSA法检测提示四种单抗抗原在肿瘤患者血清中均有分布,并对胃肠肿瘤的检出有一定帮助。外源凝集素用于单抗单抗相应抗原的分析检测是一种简便有效的工具.
Concanavalin A(Con A)and Phaseolus Vulgaris Agglutinin (PHA)were used as a tool in aiialysis of the corresponding antigens of MG monocional antibodies.The glycoprotein with N-glycosidic link was purified fiom gatric canncer tissue by Con Aaffinity chromatography, and then detected by MG monocfonal antibodies against gastriccancer It was suggested that the corresponding antigens of MGd1, MG7,MGb1 andMGcl all could be recognized by Con A, being a group of glycoproteins with N-glycosidiclink The corresponding antigens of MGdl and MG7 could also be recognized by PHA-E。A sandwjtch method was established with Con A and MG monoclonal antibody, and usedto detect these antigens in serum of patients with carcinoma.The results from this testsdemonstrate that detection of these antigens in serum of patient can be helpful for diagn-osis of gastrointestinal cancer,and that lectin can be used in isolation,analysis and detectionof the antigen defined by monoclonal antibody against carbohydrate.
